11/2/2022 0 Comments Palindromic sequence![]() Palindromes and near-palindromes also serve as binding sites for restriction enzymes and transcription factors (TFs). Because of this tendency, they are often responsible for chromosomal translocations, recombinations, and deletions and are associated with the inheritance of many genetic diseases. Palindromic AT-rich repeats (PATRRs) can cause DNA to denature more readily due to weak A–T bonds, thereby increasing the propensity of the sequence to fold into a secondary structure 11. They can alter the DNA replication process and inhibit gene expression by inhibiting ribosomal translocation along the mRNA transcript 9, 10. The sites of cruciform formation are hot spots for DNA breakage and chromosomal translocation 8. A long palindrome has the unique ability to fold back onto itself to form a secondary structure called a cruciform or hairpin (Fig. Short palindromes, under 50 bp, prevent DNA degradation, while palindromes longer than 50 bp result in mutations and DNA instability 6. They stimulate deletions during DNA replication and interchromosomal recombination between homologous sequences, leading to the loss of intervening sequences in fact, palindromes and near-palindromes account for 83% of deletions and small insertions 4, 5. Palindromes affect various cellular processes, including gene expression, regulation, and gene replication 4. Measuring the abundance, frequency, and location of palindromes across the human genome is critical to understanding their functional roles. Palindromic DNA sequences are prevalent in the genomes of a wide variety of organisms, including humans the functions and implications of such sequences are only beginning to be understood. Sequences that are palindromic except for a few mismatches in base pairing are called near-palindromes. If the complementary portions are separated by a gap sequence, the sequence is referred to as inverse repeat 3. When a palindromic sequence is folded at its midpoint, the base pairs (bp) on the two halves are complementary. 1, the sequence on the positive strand 5′-GACA|TGTC-3′ is a palindrome since GACA is followed by its complement CTGT, but appearing in reverse order as TGTC 2. In DNA, palindromes are defined as a sequence of nucleotides that are followed by its complement sequence appearing in reverse order 1. The catalog of palindromes in the reference genome and 1000 Genomes is being made available here with details on their variations in each individual genome to serve as a resource for future and retrospective whole-genome studies identifying statistically significant palindrome variations associated with diseases or traits and their roles in disease mechanisms. The analysis of disease/trait-associated single-nucleotide polymorphisms in palindromic regions showed that disease-associated risk variants are 14 times more likely to be present in palindromic regions than in other regions. We found that ~30% of the palindromes exhibit variation, some of which are caused by rare variants. In this work, we computed and analyzed the palindromic sequences in the human genome and studied their conservation in personal genomes using 1000 Genomes data. Palindromic mutations are linked to many human diseases, such as neuronal disorders, mental retardation, and various cancers. Palindromes are distributed throughout the human genome and play significant roles in gene expression and regulation. Cutting both strands of DNA harms the virus more than cutting one strand.A palindrome in DNA is like a palindrome in language, but when read backwards, it is a complement of the forward sequence effectively, the two halves of a sequence complement each other from its midpoint like in a double strand of DNA. Bacteria have evolved to 'disable' bacteria-targeting viruses (bacteriophages) using restriction enzymes that bind to these palindromic sequences. The last reason has been important in the struggle between viruses and bacteria. It is even possible that two enzymes work as a dimer to cut the palindromic sequence, further increasing efficiency. This means that the enzyme recognizes the sequence no matter from which side the enzyme approaches the DNA.Ī palindromic sequence also increases the chance that both strands of DNA are cut. A palindromic sequence is the same backwards and forwards on both sides (see image below). Luckily! because you don't want a 'pacman' that cuts DNA at random places.ĭNA is double stranded, so it has 'two sides' to which the enzyme can bind. ![]() It binds to the DNA only in one specific configuration. Enzymes such as restriction enzymes have to recognize a very specific sequence in order to carry out its task. ![]()

Bananacondas?" BWAHAHAHAHA! Man, I really need to start writing screenplays for intentionally bad movies. My biggest complaint is that Anacondas wasn't cheesy ENOUGH! If you're gonna be bad, then go as far out on the cheesy limb as you can go! Instead of a blood orchid, their research should've discovered a rare, Indonesian banana that held the secret to long life. However, this one does have more anacondas! It's been 7 years since I saw the original, but I'm pretty sure it's the better of the two. ANACONDA 3 MOVIEThe first movie had famous people in it, this one doesn't. If you're wondering how this compares to the original Anaconda, well, the two really have nothing to do with each other except for presence of an anaconda. It's funny up to a point, but eventually you start looking at your watch and thinking, "OK snake, it's time to eat this kid." The CGI looks a little goofy at times, but most of it is well done thanks to the wise decision to hide the flaws with darkness and water. We're treated to such intellectually-challenged exchanges as: "What's wrong with this picture?" "You're in it." And it's mostly dominated by Messner's Michael Paré-esque line delivery and Eugene Byrd's banshee-like screeching. And since this is a "creature feature," by definition a lot of the action takes place at night, in dark caves and in water, with only flashlights to light the way. In other words, you can expect a whole lot of stuff like a door opening slowly and then something jumping out while a loud noise is made. This is one of those movies that relies on every cliché in the B-movie book. ![]() When B-movie characters like her prance around with a huge chip on their shoulder, I usually pray for some sort of creature to sneak up and bite that chip right off. My first choice was the Jennifer Lopez wannabe. She eventually was only accenting about one word per sentence, but by the climax she started fakin' it up with reckless abandon! And I'm sure plenty of girls will think Johnny "Five O'Clock Shadow" Messner is pretty hot, but for the most part you'll spend the first 10 minutes of the movie picking which characters you want to see swallowed whole by a giant anaconda. But why do producers insist on accents being so fake? Her accent dwindled as the movie went along. My money says you really won't care about anybody involved in this production. He died soon enough, so it doesn't really matter. He looked like Gregg Rainwater from The Young Riders. Some other guy of some sort of foreign descent was thrown in there as well. But hey, we've got everything else!" And that, my friends, is your cast. Now how about somebody who's actually famous?" "Um, well, we don't exactly have that. He's in wardrobe right now having the sleeves on his tight shirt rolled up." "Perfect. And I saw that you found a white European male, but how about the protagonist?" "Oh, you mean the muscular white guy with a 2-day beard growth and husky voice? Check. She's from Georgia, but her accent sounded too realistic. ![]() How about a blonde babe with a thick, fake Southern accent?" "Check. That'll alert everybody that he's not one of those brainy Asians. "You mean the J-Lo wannabe? Check." "Good. Who is Morris Chestnut?" "Well, we thought it was a brand of chewing tobacco, but he's actually who we're gonna use as our tough black guy." "Oh, OK. Don't wanna be accused of stereotyping." "Good idea. But let's put a tough black dude in there as well. All right, I've kept this secret, but I actually sat in on a production meeting for Anacondas and here's how it went down: "Annoying black dude who just screams the whole time?" "Check. Bring the crackers because mama, this movie is bringin' the cheese! If there has ever been a more culturally diverse group of no-name actors to brave the jungles of Borneo then I'll end my movie reviewing career right now (future users of the Alan Smithee pseudonym applaud). ![]() 11/1/2022 0 Comments Dedalus and icarus![]() And to this day, when summer breezes blow and the wildflowers bloom, the voice of Perdix may still sometimes be heard, calling to his mate from among the grass and forest trees.
